Genetic diversity underpins species conservation and management goals, and describes a species' ability to adapt to changing environments. Using environmental DNA (eDNA) metabarcoding, we examined mitochondrial genetic diversity using multi gene sequence data of four Oncorhynchus species across 16 sites in Oregon and northern California. Our multi gene panel includes three targets commonly used in population genetic (ND2), phylogenetic (COI) and eDNA (12S ribosomal DNA) screening. The characterization of diversity at multiple loci expands the knowledge gained from eDNA sampling and provides comprehensive information about genetic diversity for management and conservation actions.
