The terminal 0-5 mm root segments of B73 seedling roots were excised, fixed in 1% formaldehyde for 15 minutes, washed in PBS, and snap frozen. Chromatin was extracted from frozen roots and used for ChIP with an anti-H3K56ac antibody (Millipore, 07-677, lot DAM1462569) and the resulting DNA Illumina sequenced.
