16S rRNA gene pyrosequencing and miseq sequencing and metagenomic sequencing were performed for deep granitic groundwater to profile microbial communities and reconstruct metabolism of subsurface anaerobic methanotrophic archaea. High-quality groundwater was collected from underground boreholes drilled at a 300-m deep stage of the Mizunami Underground Research Laboratory in central Japan.
