Setd2 is a histone H3K36 methyltransferase that suppresses lung cancer growth. Here we CRISPR knock out Setd2 in Kras-G12D genetically-engineered mouse models, and perform RNA-seq on FACS-purified neoplastic cells to profile the gene expression changes that result from Setd2 inactivation. Control group samples were transduced with inert sgRNAs. H3K36me3 immunoblot was performed to verify Setd2 knockout status. Kras-LSL-G12D;Rosa26-LSL-tdTomato;H11-LSL-Cas9 mice were transduced with Lenti-sgSetd2/Cre or Lenti-sgInert/Cre control to initiate lung neoplasia. Tumors were allowed to grow for 9-12 months before tdTomato+/CD31-/CD45-/Ter119-/F4/80- neoplastic cells were FACS purified for RNA-seq.
