Hypothesis: Vitreous protects neurons from hemoglobin (Hb) toxicity. Model: Primary cortical cell cultures (murine); cultures treated with Hb 10 micromolar alone or with bovine vitreous or vitreous components. Outcome measures: Lactate dehydrogenase (LDH)release, propidium iodide fluorescence staining intensity, culture malondialdehyde (MDA), culture iron.
研究假说:玻璃体(vitreous)可保护神经元免受血红蛋白(hemoglobin, Hb)毒性损伤。实验模型:小鼠源性原代皮层细胞培养物;将该细胞培养物分别以10微摩尔浓度的血红蛋白单独处理,或联合牛玻璃体或玻璃体组分进行处理。结局指标:乳酸脱氢酶(lactate dehydrogenase, LDH)释放量、碘化丙啶(propidium iodide)荧光染色强度、培养物丙二醛(malondialdehyde, MDA)含量、培养物中铁含量。
