To investigate the possible impact of MOK-inhibition in early transcriptional responses induced by TDP-43 aggregates, we tested the effect of AG2P145D/C13-pretreatment of SIM-A9 cells and subsequent exposure to TDP-43 (or sham) aggregates. Comparative transcriptional analysis of (nascent chain) SLAM-Seq data for SIM-A9 cells in culture treated with polymixin B for 1 h and then either with 10 uM AG2P145D/C13 (Garske et al., PNAS 2011; doi: 10.1073/pnas.1111239108) or DMSO for 1h. Subsequently, TDP-43- or sham-aggregates were added to 5 ug/mL final concentration (2.5 h), followed by addition of 4-thiouridine (S4U) to 100 uM final concentration and further incubation for a further 90 min.
