Whole-genome sequencing of Ligilactobacillus salivarius GHL01 derived from poultry lungs

This study aimed to screen poultry lung-derived Lactobacillus strains with effective antibacterial properties. Lung tissues were aseptically collected from high-performance broiler chickens (43-day-old male chickens weighing 3.5+/-0.1kg, n:10) sourced from a large-scale commercial broiler farm, and pooled. The tissues were homogenized, and the homogenate was eluted with sterile pre-cooled phosphate-buffered saline (PBS) into sterile MRS broth for enrichment and plating. Simultaneously, direct plating on MRS agar was performed to isolate single colonies for purification. Subsequent analyses included morphological identification, antibacterial activity assessment, antibiotic susceptibility testing, acid-producing capacity evaluation, and acid and salt tolerance tests. A strain exhibiting superior performance was selected and designated as Ligilactobacillus salivarius GHL01. Whole-genome sequencing of this strain was conducted using a combination of second-generation Illumina sequencing and third-generation Nanopore sequencing.
