Identifying novel therapeutics for subjects with idiopathicpulmonary fibrosis (IPF), a progressive and fatal disorder without curative therapies, is a critical unmet need. eNAMPT (extracellular nicotinamide phosphoribosyltransferase) is a damage-associated molecular pattern protein (DAMP), and Toll-Like Receptor 4(TLR4) ligand contributing to disease severity in preclinical radiation-induced lungfibrosis and NASH-associated hepatic fibrosis. Using scRNA sequencing from a preclinical intratracheal bleomycin-exposed model of lung fibrosis in C57l6 mice, we examined eNAMPT as a druggable target in human IPF and preclinical PF, utilizing ALT-100, a humanized eNAMPT- neutralizing mAb.
