Metagenomic Analysis of Bacteria and Archaea in Landfill Bioreactor

The purpose of this project is to show how pilot scale bioreactors containing sewage sludge mixed with municipal solid waste (MSW) and a geotextile filter in its drainage layer can improve leachate quality and waste stabilization and the diversity of microorganisms accumulated on the geotextile fabrics investigated. After 540 days of operation, each geotextile fabric was removed from the reactors and air-dried over a night. Then, two 1 cm x 1 cm geotextile specimens were cut from each sample for microbial analysis. Geotextile samples extracted from the bioreactors at the end of the operation were cut in 1cm2 size in a sterile environment on the same day and samples were transferred to eppendorf tubes filled with Glycerol. DNA was extracted from the samples using the DNeasy PowerBiofilm Kit.
