Brucella proline racemase protein A targets Tpl2 to promote IL-10 secretion for establishment of chronic infection

This dataset includes the IL-10 concentration in macrophages after stimulation with rRomA, rOMP25, rwadC, and rPrpA proteins in vitro and in vivo. It also includes the IL-10 concentration in macrophages aftertransfection with varying doses of the pcDNA3.1-prpA plasmid. All IL-10 concentrations were determined using ELISA kits. Additionally, the dataset contains data on the number of cells induced by PrpA, as well as the proliferation rate of macrophages (evaluated using CCK-8 kits). Furthermore, the dataset includes data on the intracellular survival of M5-90, the M5-90 prpA mutant, and M5-90 prpA-c in murine macrophages. It also comprises proteomics data of murine macrophages infected with M5-90 and the M5-90 prpA mutant, along with results from molecular dynamics simulations of the PrpA and Tpl2 proteins. Finally, the dataset includes the following: 1) Spleen colony-forming units (CFUs) of mice inoculated with M5-90 and the M5-90 prpA mutant at 30 days post-inoculation; 2) Serum levels of Brucella-specific IgG antibody, IFN-γ, TNF-α, and IL-10 (evaluated using ELISA kits) at 30 and 45 days post-inoculation; 3) Flow cytometry analysis data of splenic lymphocytes from mice immunized with M5-90 and the M5-90 prpA mutant at 30 and 45 days post-immunization; 4) CFU counts in the spleen from challenge experiments.
