Purpose of this project is to understand the molecular mechanism of taste stem cell maintenance and taste cell differentiation using organoid culture system. We have isolated Lgr5-GFP taste stem/progenitor cells from circumvallate papillae and cultured them in vitro. All different types of taste cells can be found in our in vitro 3-D culture system after 12-14 days of culture. With RNAs extracted from Day 2 (D2), D4, D6, D8, D10, D12 and D14 taste organoids, we have systematically surveyed the transcriptome landscape of taste organoids at different developmental stages using RNA-seq.
