scRNA-Seq MP-CD8 Ly49+ following viral infection

Based on CD8b expression we have defined two MP-Ly49+ CD8 cells subsets. To characterize the activation of MP-Ly49+ CD8 cells during the course of an antiviral response, we performed a single-cell transcriptomic analysis. We sorted both CD8aa and CD8ab-Ly49+ cells from the spleen of naive or VV-infected mice at 8 days post-infection. We also sorted Ly49-CD44+ and naive cells from naive mice and B8R-specific cells from VV-infected mice, as controls.基于CD8β（CD8b）的表达特征，我们定义了两个MP-Ly49阳性CD8 T细胞亚群。为了表征抗病毒应答过程中MP-Ly49阳性CD8 T细胞的激活状态，我们开展了单细胞转录组分析（single-cell transcriptomic analysis）。我们从未致敏小鼠的脾脏，以及感染后第8天的痘苗病毒（Vaccinia Virus, VV）感染小鼠的脾脏中，分选出CD8αα（CD8aa）与CD8αβ（CD8ab）-Ly49阳性细胞。同时，我们从未致敏小鼠体内分选出Ly49阴性CD44阳性细胞与未致敏细胞，并从VV感染小鼠体内分选出B8R特异性细胞作为对照。
