MCF7 cell line transcriptome after cisplatin and sip53

In this study we compared transcriptomes of MCF7 cells treated with cisplatin (5 uM) and with transient deficiency of p53, which was silenced with siRNA. Total RNA was isolated with TRI Reagent, mRNA was isolated using NEBNext Poly(A) mRNA Magnetic Isolation Module. Next mRNA was converted do cDNA using NEBNext Ultra II RNA First Strand Synthesis Module and NEBNext Ultra II Non-Directional RNA Second Strand Module. The purified cDNA was used for RNAseq library preparation using MGIEasy PCR Free DNA Library Prep Set according to the protocol. Sequencing was performed using DNBSEQ G400.
