Type-VI glandular trichomes of wild tomato Solanum habrochaites PI127826 produce high levels of the sesquiterpene 7-epizingiberene and its derivatives, making the plant both repellent and toxic to several pest insects and pathogens. How wild tomato trichomes achieve such high terpene production is still largely unknown. A cross (F1) between the a cultivated tomato S. lycopersicum and S. habrochaites PI127826 was generated that produced only minute levels of 7-epizingiberene. The F2 progeny, selected for the presence of the 7-epizingiberene biosynthesis genes, was backcrossed to the cultivated tomato parent to generate a BC1F3. One BC1F3 line with the âhigh-production active-trichome phenotypeâ of PI127826 and one BC1F3 with the "low-production lazy-trichome phenotype" of the parental cultivar were selected for sequencing. The wild parent PI127826 was also sequenced. Trichome type VI glandular heads were isolated, their total RNA isolated and the corresponding mRNA sequenced. This study is part of the same experiment than study with the EBI ENA accession PRJEB58757.
