Deep sequencing of cDNA of Neisseria gonorrhoeae bacteria that were grown in suspension or adherent to acid-washed glass coverslips, in media where zinc was sequestered or available in excess. Total RNA was isolated from Neisseria gonorrhoeae strain FA1090 grown for four hours either in suspension or adherent to acid-washed glass coverslips. Three media conditions were tested: Keratinocyte serum-free medium (KSFM) without additives, KSFM with 3 μM recombinant murine calpotectin, or KSFM with 3 μM mouse calprotectin and 6 μM ZnSO4. Ribosomal RNA was depleted, and remaining RNA was reverse-transcribed into a cDNA library for Illumina sequencing. Reads were mapped to the FA1090 genome.
