Effects of hindlimb ischemia on bone marrow leukocytes and hematopoietic stem and progenitor cells

We identified the genes regulated by hindlimb ischemia in leukocytes and hematopoietic stem and progenitor cells isolated from the bone marrow in mice. We selected some of these genes based on their expression and further evaluated their functions using a wise array of techniques. Overall design: Hindlimb ischemia (HI) was performed in mice, and leukocytes and hematopoietic stem and progenior cells (HSPC) were sorted from the bone marrow on days 2 and 7 after HI. mRNA was extracted from these samples and single cell RNA sequencing was performed.
