To explore the cellular heterogeneity in the developing and functionally mature thymus, we constructed an integrative transcriptomic atlas of human thymocytes involving both prenatal and postnatal stages. Specifically, we characterized the following aspects: (i) defining the prenatal DN subsets transcriptomically, (ii) revealing the transcriptional differences between prenatal and postnatal DN cells regarding key genes expression, enriched GO terms as well as metabolic programs, and (iii) illustrating the different preference of polyadenylation sites usage in prenatal and postnatal thymocytes at single-cell resolution. Supplementary tables and datasets include the re-processed datasets, integrated annotation of each cell cluster and subcluster, UMAP dimensionality reduction, scaled regulon activities, pesudotime inferred by Monocle 3, and patterns genes.
为探究发育中与功能成熟胸腺内的细胞异质性,我们构建了涵盖产前及产后阶段的人胸腺细胞整合式转录组图谱。具体而言,我们从以下维度开展表征:(i) 从转录组层面定义产前双阴性(double negative, DN)细胞亚群;(ii) 揭示产前与产后双阴性细胞在关键基因表达、富集的基因本体(Gene Ontology, GO)条目以及代谢程序上的转录差异;(iii) 在单细胞分辨率下阐明产前与产后胸腺细胞在多聚腺苷酸化位点(polyadenylation site)使用偏好上的差异。本研究附带的补充表格与数据集包含:经重新处理的数据集、各细胞簇与亚簇的整合注释、均匀流形近似与投影(Uniform Manifold Approximation and Projection, UMAP)降维结果、标准化调控子活性、由Monocle 3推导的拟时间,以及特征基因表达模式。
