Single-molecule force spectroscopy data for prion-like conversion of wild-type SOD1 by pathogenic mutants G127X and G85R

Excel files containing (A) force-extension curve data and (B) ensemble assay data for studies on prion-like propagation of misfolding in SOD1.(A) Force-extension curve data from optical tweezers measurements of the unfolding and refolding of: (i) wild-type (WT)-G127X heterodimers of superoxide dismutase-1 (SOD1), pulling on the termini of the WT domain; (ii) WT-G127X SOD1 heterodimers pulling on the termini of the G127X domain; (iii) WT-G85R SOD1 heterodimers, pulling on the termini of the WT domain; and (iv) WT-WT homodimers (control), pulling on the termini of one of the WT domains. The file title indicates which construct was used in the measurements. In each file, data are presented as paired columns of force ("F", values listed in pN) and extension ("Ext", values listed in nm) data for sequential unfolding and refolding measurements (refolding data indicated by "_r" in column titles). The remainder of the column name indicates the molecule number (molecule 1, 2,...), followed by an underscore and the number for the set of curves measured from that molecule (set 1, 2,...), followed by an underscore and the number of the pull within that set (pull 1, 2,...). Each set of FECs is organized in a separate sheet in each file, with the tabs for the sheets labelled in the format MoleculeNumber_SetNumber.(B) Data from enzymatic assays and mass photometry assays of prion-like conversion in SOD1. Conversion assay results show the percent inhibition of WST-1 formazan formation by SOD1 at different time-points and under different solution conditions. Mass photometry results list the masses for all binding and unbinding events (respectively, positive and negative values) with SOD1 heterodimer alone, 10C12 antibody alone, and SOD1+antibody, at 0 and 48 hr incubation times.
