scRNAseq of diabetic corneal epithelium

We performed single cell transcriptional profiling of mouse corneal epithelium in streptozotocin-induced diabetic mice. We prepared cell suspensions of corneal epithelium: one from control and one from diabetic C57BL/6J mice (male, 20 weeks old). Each sample is a pooled sample of 4 corneas from n = 2 mice. We induced hyperglycemia by administering a single injection of streptozotocin (STZ, 160 mg/kg bodyweight) to cause complete pancreatic β-cell necrosis and diabetes, with control mice receiving an equal volume of sodium citrate buffer (pH 4.5). We measured blood glucose weekly to certify hyperglycemia, and mice that remained hyperglycemic (blood glucose > 250 mg/dL) throughout the 12 week study were considered diabetic.
