The global rise in antimicrobial resistance (AMR), particularly among WHO-priority pathogens like Escherichia coli and Klebsiella pneumoniae, is significantly driven by the horizontal transfer of beta-lactamase genes via plasmids. This project aims to identify conserved plasmid segments carrying mismatch repair genes, mobile genetic elements, and beta-lactamase resistance variants (blaVIM, blaOXA-48, blaKPC) across E. coli and K. pneumoniae isolates, using Vivli AMR surveillance datasets. Through an integrative genomics approach involving plasmid reconstruction, comparative genomics, and phylogenetics, the project will characterize key plasmid features promoting resistance dissemination. Expected outcomes include the identification of conserved plasmid backbones, insights into genetic contexts of beta-lactamase genes, and a deeper understanding of plasmid-driven AMR spread. Findings will have important implications for AMR surveillance, informing targeted interventions to curb plasmid-mediated resistance.By addressing these gaps, this project seeks to illuminate plasmid-mediated AMR pathways, enabling more effective containment strategies.
