Curcumol Protects Against Myocardial Infarction by targeting STING to Suppress Type I Interferon Response in Macrophage

Macrophages play a crucial role in the pathological process after myocardial infarction (MI). Curcumol (Cur), a natural compound extracted from the Curcumae Rhizoma (CR), has demonstrated both anti-tumor and anti-inflammatory activities. We had found that Cur had cardio-protective effect in MI and the effect was reduced thanks to macrophage depletion. To elucidate the mechanism of Cur on macrophage in MI, we performed RNA-seq on macrophages in MI heart treated with Cur or not. MI model were performed on C57BL/6JGpt male mice. In curcumol treatement group, mice were treated with 30 mg/kg curcumol was administered via oral gavage once daily for 7 days. CD45+CD64+ macrophages were isolated by FACS from hearts of mice treated with or without curcumol on day 7 after MI. RNA integrity was assessed using the Agilent 2100 Bioanalyzer (Agilent Technologies, Santa Clara). Then the libraries were constructed using TruSeq Stranded mRNA LT Sample Prep Kit (Illumina, USA) according to the manufacturer’s instructions.
