Dataset on proteomics in SH-SY5Y cells after leucine treatment

In this dataset, an integrated method of drug affinity responsive target stability (DARTS) and Astral DIA quantitative proteomics was used to screen the direct protein targets of leucine. SH-SY5Y cells were washed with PBS, lysed on ice for 15 min using the lysis buffer, and incubated with leucine. The protein mixture was separated in SDS-PAGE and stained with Coomassie brilliant blue dye. The target protein bands were excised from the gel and subjected to Astral DIA quantitative proteomics analysis.本数据集采用药物亲和力响应靶标稳定性（drug affinity responsive target stability, DARTS）与Astral DIA定量蛋白质组学的整合方法，筛选亮氨酸的直接蛋白质靶标。具体实验步骤如下：将SH-SY5Y细胞用磷酸盐缓冲液（Phosphate Buffered Saline, PBS）洗涤后，于冰上使用裂解缓冲液裂解15分钟，并与亮氨酸共孵育；随后将蛋白质混合物通过十二烷基硫酸钠-聚丙烯酰胺凝胶电泳（SDS-PAGE）分离，并用考马斯亮蓝染料进行染色；最后从凝胶中切取目标蛋白条带，开展Astral DIA定量蛋白质组学分析。
