MAP6 is a neuronal microtubule inner protein that stabilizes microtubules. By cryo-ET, using purified tubulins and actins, we recently confirmed that MAP6 is able to organize actin filaments in the lumen of microtubules in vitro. For that purpose, we over-expressed MAP6 in mouse neurons that grow directly on EM grids. Our goal is therefore to observe this kind of architecture in young neurons and more specifically in their growth cones as there are enriched in actin and rather flat (so more easily to be imaged by EM) using cryo-electron tomography.
