We engineered a new m6A editing system consisting of GCN4 (SunTag)-dCas13b, anti-GCN4 single chain variable fragment (scFv)-FTO or ALKBH5-GBI and sgRNA to provide an applicable method to remove m6A modification at specific loci. Overall design: m6A-seq were performed to evaluate the demethylation results of targeted SON demethylation samples (WT (sg-SON)), off-target control (WT (sg-control)) and mutant FTO control (Mut (sg-SON)). RNA lifetime profiling was conducted by collecting and analyzing RNA-seq data on targeted SON demethylation and control samples, the transcription of which were inhibited with actinomycin D at different time points.
