Whole-exome sequencing of naive and Ara-C resistant KG-1a

To ascertain whether exomic mutations play a facilitative role in the development of Ara-C resistance in KG-1a cells, we undertook exomic sequencing on both naive and Ara-C resistant KG-1a cells. Our analysis identified 297 mutations, with the majority being intronic (41.8%), followed by intergenic (21.9%), and a smaller proportion being exonic (12.8%) . A total of 21 nonsynonymous mutations were measured across 20 genes. Among these, ZNF148 and ZNF827, which are implicated in the regulation of gene expression, and RRM1, a gene critical for DNA replication during the S phase of the cell cycle and involved in various DNA repair processes, were highlighted. However, our study did not uncover any nonsynonymous mutations within genes that have been previously reported to be directly associated with drug resistance. These findings suggest that, in the short term, exomic mutations may not substantially contribute to the development of Ara-C resistance in KG-1a cells.
