In this setup we have used RNA-seq, ChIP-seq, and ATAC-seq to identify how natural mutations in PPARg, which leads to lipodystrophy, cause changes in transcription, chromatin landscape, transcription factor binding and enhancer activity. Overall design: Global gene expression analysis, HA-PPARg ChIP-seq, Med1 ChIP-seq, H3K27ac ChIP-seq, and ATAC-seq in PPARg-/- MEF-coxsackievirus-adenovirus receptor (CAR) cells acutely transduced with adenovirus expressing PPARg2-WT, PPARg2-E379K, or PPARg2-R212Q. All experiments are performed in duplicates.
