A CRISPR-Cas9 loss-of-function library GeCKO 2.0 based on whole-population mRNA was transfected in the HNSCC primary cell line Fadu and metastasis cell line Detroit 562 for loss of function screening to understand the functions and pathways involved in the drug response in HNSCCs. Repeated screening was performed using cisplatin or 5-fluorouracil, and the related drug resistance candidate genes were narrowed to 6,848 genes.
