Source data for lineage- and program-selective correspondence between canine chronic inflammatory enteropathy and human Crohn disease
收藏资源简介:
This repository provides the processed source data supporting the study entitled “Lineage- and program-selective correspondence between canine chronic inflammatory enteropathy and human Crohn disease.” The study investigates disease-associated transcriptional changes across canine chronic inflammatory enteropathy (CIE) and human Crohn disease using publicly available single-cell RNA sequencing datasets. Analyses were conducted at the donor or participant level, focusing on epithelial cells, T cells, and myeloid cells. Cross-species comparisons were evaluated at the gene, transcriptional program, and cell-state levels, with additional sensitivity analyses addressing donor stability, anatomical region, disease context, and technical variation. Data sources The analyses were based on the following publicly available Gene Expression Omnibus (GEO) datasets: GSE254005:Canine CIE discovery cohort, comprising three healthy dogs and four dogs with CIE.GSE266616: Human terminal ileum single-cell dataset, including the primary comparison of 16 healthy participants and 11 participants with active Crohn disease.GSE185224:Healthy duodenum and terminal ileum samples from three paired donors, used to evaluate anatomical-region effects.GSE277276: Human coeliac disease dataset, including 11 healthy controls and 21 active cases, used to assess disease-context specificity. Repository contents The deposited files include: SourceData.zip: Numerical source data underlying six main figures and five supplementary figures, including gene-level and pathway-level analyses, cross-species comparisons, cell-state analyses, and sensitivity assessments.SupplementaryData01.xlsx: Complete evidence matrix for 25 canine-derived lineage–program units, including cross-species evidence, disease-context information, final evidence categories, and confidence classifications.SupplementaryData02.xlsx: Complete leave-one-dog-out (LODO) stability results for the 25 predefined canine lineage–program units. README.md:Documentation describing the deposited files, their organization, and their relationship to the figures and analyses. Data interpretation and reuse These materials provide traceable numerical results supporting the figures and supplementary analyses. The 25-program evidence classifications represent an exploratory, rule-based synthesis of multiple evidence sources; they do not constitute independently validated mechanisms or a single confirmatory false discovery rate testing framework. The deposited files contain processed analysis outputs rather than redistributed raw single-cell sequencing matrices. Original sequencing data remain available through the GEO accession numbers listed above. The dataset is intended to support transparency, independent evaluation, reproducibility, and further comparative research into canine and human intestinal inflammatory diseases.
