<p>Fluorescence was measured over 24-48 hours incubation time with a plate reader (TECAN spectrafluor plus; 360 nm excitation, 460 emission), with time points taken every 4-6 hours. Hydrolysis rates were calculated from the rate of increase of fluorescence in the incubation over time relative to a set of standards of known concentration of fluorophore. Scripts to calculate hydrolysis rates and produce the figures shown here are available in the associated Github repository [Hoarfrost, 2017].</p>

本研究采用TECAN Spectrafluor Plus型酶标仪(激发波长360 nm,发射波长460 nm),在24~48小时的孵育周期内检测荧光信号,每4~6小时采集一组时间点数据。水解速率通过孵育体系中荧光信号随时间的增长速率,结合一系列已知浓度的荧光团(fluorophore)标准品计算得到。用于计算水解速率及生成本文附图的代码脚本,可在关联的GitHub代码仓库[Hoarfrost, 2017]中获取。
