Genomic DNA from 10 different strains of Thermus thermophilus was prepared and mixed in equimolar amounts for sequencing. The 10 strains were selected on kanamycin after transformation of a Thermus thermophilus Primpol gene deletion mutant with Thermus thermophilus genomic DNA that has been subject to a transposition reaction with a Tn5 transposase and a transposable fragment that contains a kanamycin resistance cassette. The purpose of the sequencing is to verify the transposition events.
