Nanostring GeoMx DSP data for TNBC Samples_ M-712Baylor

Gene expression analysis using the GeoMX DSP system (NanoString) was conducted to profile 57 FFPE TNBC tissues from self-reported 26 BA and 31 WA patients. We examined the transcriptome of these samples using the GeoMX Human Whole Transcriptome Atlas (WTA), which measures approximately 18,000 protein-coding genes. The GeoMX DSP analysis involved staining the slides with two morphology markers—Pan Cytokeratin (PanCK) to identify tumor cells, and CD45 to identify immune cells—along with a nuclear stain (DAPI).本研究采用GeoMX数字空间解析系统（GeoMX DSP, NanoString）开展基因表达分析，对26名自我报告为BA、31名自我报告为WA的患者的57份福尔马林固定石蜡包埋（FFPE）三阴性乳腺癌（TNBC）组织样本进行转录组谱分析。我们采用GeoMX人类全转录组图谱（WTA）对上述样本的转录组进行检测，该图谱可覆盖约18000个蛋白质编码基因的表达量分析。GeoMX DSP分析流程包括使用两种形态学标记物对组织切片进行染色：以细胞角蛋白广谱抗体（Pan Cytokeratin, PanCK）标记肿瘤细胞、以CD45标记免疫细胞，同时辅以细胞核染料DAPI进行核染色。
