Expression data from mRNAs co-precipitated with QKI in SW480 cells

In CRC cells, lncRNA NONHSAT136151 can bind to protein QKI but does not affect the expression of QKI. The binding of NONHSAT136151 may change the interactino of QKI with its target genes. We used microarrays to detail the abundance of mRNAs enriched by QKI in NONHSAT136151 silenced group and control group in CRC cells. SW480 cells treated with siLnc (siNONHSAT136151) and siNC (Negative control) were lysed with RIP lysis buffer. Anti-QKI antibody or IgG from rabbit serum were pre-bound to Protein A Magnetic Beads and then incubated with cell lysate at 4°C overnight with rotation. The magnetic bead-antibody-QKI complex was washed and resuspended with RIP wash buffer. RNA was recovered using TRI Reagent and dissolved in RNAse-free water. Subsequently, PrimeViewTM Human Gene Expression Array (Affymetrix Inc.) was utilized to analyze the RNA samples.
