Porcine B-cell repertoires against foot-and-mouth disease virus

Excavation of inter-serotype broadly neutralizing neutralization antibody (bnAb) was challenging for foot-and-mouth disease virus (FMDV), due to its existence as seven distinct serotypes without cross-protection. Here we designed an elegant strategy for the precise mining of bnAbs by construction and dissection of different antigen-specific B cell receptors (BCRs) repertoires derived from the same animal. The FMDV serotype O and A specific class-switched B cells were respectively purified from PBMCs in the vaccinated pig and submitted to high-throughput transcript sequencing for production of BCR repertoires. Our study aimed to provide an elegant strategy to uncover bnAbs for highly mutable pathogens as well as cross-serotype antigen structures to explore of broad-protection FMDV vaccine.
