The experiments carried out to identify and quantify the various proteins extracted from Aedes aegypti mosquito sample upon CHIKV and DENV mono- and co-infection at 24 hrs post infection. Proteins from the tissue samples were extracted, reduced, alkylated and digested with trypsin. To obtain quantitative information, the peptides were labelled with TMT isobaric tags, mixed, fractionated and analysed by mass spectrometry. The proteins thus obtained were then subjected to basic bio-informatics analysis.
本实验旨在鉴定并定量分析感染后24小时的埃及伊蚊(Aedes aegypti)样本在基孔肯雅病毒(CHIKV)、登革病毒(DENV)单一感染及共感染条件下提取的各类蛋白质。研究人员首先对该组织样本中的蛋白质进行提取、还原、烷基化处理,并以胰蛋白酶进行酶解;为获取定量信息,将所得肽段用TMT同量异位素标签(Tandem Mass Tag)进行标记,随后混合、分级分离并通过质谱分析法开展检测分析;最终获得的蛋白质组将进一步进行基础生物信息学分析。
