Given that [beta]-catenin is a well-known transcriptional coactivator, and its transcriptional downstream genes may mediate [beta]-catenin’s action on hepatocarcinogenesis. Here, we performed ChIP-seq to study genome-wide distribution of [beta]-catenin in [beta]-catenin[delta](ex3)/+ livers in quest of the putative genes. Three [beta]-catenin[delta](ex3)/+ mouse livers were used for [beta]-catenin ChIP-seq. Livers were sonicated using a M220 Focused-ultrasonicator (Covaris, Woburn, MA, USA). Chromatin was immunoprecipitated with 4μg antibodies against [beta]-catenin (Carboxy-terminal Antigen, #9587S) overnight at 4°C. ChIP-Seq libraries were prepared using Kapa LTP Library Preparation Kit (Kapa Biosystems, Sigma-Aldrich). ChIP-Seq was performed on Illumina HiSeq3000. For the bioinformatic analysis of ChIP-Seq data, Bowtie2 was used for alignment. Enriched peaks were identified using MACS2 with FDR < 0.01. ChIPseeker was used for peaks annotation.
