This case-control group includes 488 cases of cervical cancer, 684 cases of cervical precancerous lesions, and 510 healthy females. Multiplex polymerase chain reaction (PCR) combined with next-generation sequencing method was used for determination of gene polymorphisms (rs5498, rs3093030 and rs281432). In our study, we divide cervical cancer into two subgroups: cervical squamous cell carcinoma (CSCC) group and cervical adenocarcinoma (CAC) group. We analyzed alleles and genotypes of all research subjects, and adjusted the age using binary logistic regression analysis. In addition, we also analyzed the distribution of haplotypes of the three SNPs in cervical cancer and cervical precancerous lesions. We found that rs3093030 can reduce the risk of cervical cancer, and rs5498 has a protective effect on the susceptibility of CAC. In addition, the frequency of haplotype "CTG" was significantly lower in the CAC group than in the controls. In conclusion, the study suggested that <i>ICAM-1</i> gene polymorphisms may have a potential role in the pathogenesis of cervical cancer in the northern Chinese han population.
本病例对照研究队列共纳入488例宫颈癌患者、684例宫颈癌前病变患者及510名健康女性。本研究采用多重聚合酶链反应(PCR)联合下一代测序技术,对3个基因多态性位点(rs5498、rs3093030及rs281432)进行分型检测。本研究将宫颈癌患者分为两个亚组:宫颈鳞状细胞癌(CSCC)组与宫颈腺癌(CAC)组。对所有研究对象的等位基因与基因型进行分析,并采用二元logistic回归校正年龄因素。此外,本研究还分析了3个单核苷酸多态性(SNPs)位点的单倍型在宫颈癌患者与宫颈癌前病变患者中的分布情况。研究结果显示,rs3093030可降低宫颈癌的发病风险,而rs5498对宫颈腺癌(CAC)的易感性具有保护作用。此外,宫颈腺癌(CAC)组中单倍型"CTG"的频率显著低于对照组。综上,本研究提示,细胞间黏附分子1(ICAM-1)基因多态性可能在中国北方汉族人群宫颈癌的发病机制中发挥潜在作用。
