Background mutagenesis during prolonged exposure

Assessment of accumulation of background mutations in HepG2 and MCF10A during 2 months of culture.Per cell line (MCF10A or HepG2) 3 single cell clones were established. MCF10A_SC7 and HepG2_SC1 were then subjected to a mock exposure protocol, consisting of 2 months of continuous culture followed by single cell sorting to generate '2nd generation clones' HepG2_SC1_cl1-3 and MCF10A_SC7_cl1-3. WGS was performed on all of these clones to establish the background mutagenesis levels and mutation spectrum in these cell lines
