Transcriptome sequencing is a useful approach for quantifying gene expression and for establishing coding sequences. This is especially true in species without an available reference genome sequence. Recent successful applications of RNA sequencing in conjunction with de novo transcriptome assembly have enabled the identification of novel genes and new behavioral patterns. Here, we reconstructed a de novo transcriptome of the Hirudo Medicinalis central nervous system (CNS). We analyzed transcription levels in three locations along the central nervous system. Coupling location and transcription in the CNS provides a first of a kind spatial regulation map of the Hirudo medicinalis CNS. By identifying unique groups of genes, according to their differential expression along the CNS, we facilitate an additional levels of understanding of gene functions. This level of understanding is tethered to a host of neurological conditions, in which the presented phenotype is dependent on neuronal locale.
