Library construction and genome sequencing of S. reilianum strain SRZ2 was done at 454 Life Sciences (USA). As a first step a whole genome shotgun random library was constructed and sequenced using the Genome Sequencer 20 (Roche). In a second step, the genome was sequenced from a newly generated paired-end library of S. reilianum strain SRZ2 using the Genome Sequencer FLX (Roche). In a third step, the scaffold-assembly was verified by generating an NheI-based Optical Map (OpGen). Optical Mapping resulted in the assembly of 23 chromosomes on which 99.75% of the genome assembly could be placed. The total genome size based on Optical Mapping is estimated to be 18.7 Mb. Thus, the assembly contains 97 % of total sequence information.
