Improved Workflows for High Throughput Library Preparation using the Transposome-Based Nextera System

We have modified the Nextera transposome-based approach for next-generation sequencing library production, removing several limitations of the original protocol. Firstly, we introduced an initial bead-based normalisation step which also improves sample purity. Secondly, we reduced reagent volumes to bring the cost into line with our existing library preparation protocol. Lastly we enabled higher throughput by introducing bead-based clean-up and a larger, custom set of index tags. We tested the robustness of the modified kit across genomes with a wide range of GC contents.
