Citrus Huanglongbing (HLB) disease, caused by Candidatus Liberibacter asiaticus (Las), is the most devastating disease in the world citrus industry. Salicylic acid plays a central role in regulating plant defense to pathogen attack. Salicylic acid (SA) methyltransferase (SAMT) modulates SA homeostasis by converting SA to methyl salicylate (MeSA). Overexpression of CsSAMT1 in Wanjincheng orange significantly enhanced tolerance to Las. RNA-seq analysis revealed that overexpression of CsSAMT1 upregulated transcriptional activities of defense genes, especially, TIR-NBS-LRR disease resistance protein family involving in plant SA and MeSA-mediated basic resistance. This study provides new insights for improving host resistance to HLB by manipulation of SA and MeSA signaling in citrus.
