Nucleosome is the fundamental structural unit of chromatin and plays a critical role in regulation of gene expression. Nucleosome occupancy is reorganized during somatic cell reprogramming, but how nucleosome positioning is changed during reprogramming remains poorly understood. Here, we established a method for depicting genome-wide nucleosome maps of porcine embryonic fibroblasts (PEFs) and pluripotent cells reprogrammed from PEFs using somatic cell nuclear transfer method (SCNT) with as low as 1000 cells using MNase-seq to find the nucleosome changing in the process of reprogramming.
