Dendritic cells (DCs) simultaneously prime and dampen the immune response. The transcriptional program that regulates immunosuppression in DCs is currently unknown. Here we identified ETS homologous factor as a key transcription factor that regulates DC immunosuppression. Mice were immunized subcutaneously (s.c.) in the rear footpads with or without 10 µg TLR stimulation in 40 µL PBS. Popliteal lymph node cells were collected 48 hrs after TLR stimulation for cDCs (live MHCII+ CD11c+) FACS-sorting for scRNA-seq library preparation.
