Cirrhosis is a prevalent global health issue that imposes a significant economic burden. The lack of effective treatments is attributed to the persistent inflammation within the liver, uncontrolled accumulation of reactive oxygen species (ROS), and poor patient compliance with injectable medications. Here, we reported that tFNA can be effectively absorbed into the blood through jejunum injection and delivered to macrophages in the liver. We developed BiRDS consisting of tFNA and a targeted siRNA against galecitin 3 to inhibit macrophage M1 polarization and inflammatory responses. RNA-sequecing was employed to explore the mechanisms of BiRDS on LPS and IFN-r treated RAW264.7.
