This study was designed to investigate the molecular response mechanisms of the mulberry looper, Phthonandria atrilineata, a significant agricultural pest, to chemical insecticides with distinct modes of action using RNA-Seq technology. Third-instar larvae were treated with sub-lethal concentrations of several insecticides, including the organophosphates trichlorfon and phoxim, the mitochondrial inhibitors chlorfenapyr and tolfenpyrad, and a trichlorfon-malathion mixture, alongside an untreated control group. Surviving larvae were collected 48 hours post-treatment, and total RNA was extracted for high-throughput sequencing on an Illumina NovaSeq 6000 platform. This dataset contains the raw sequencing reads from all treatment and control groups and serves as a valuable resource for elucidating the general and specific gene expression regulatory networks in P. atrilineata under various chemical stresses, aiming to provide insights into the molecular basis of its detoxification metabolism, insecticide resistance, and stress adaptation.
