Transcriptional profiling of livers in Apoa5 knockdown mice

Sepsis-associated liver injury (SALI) is associated with high mortality and abnormal immune cells activation. Apolipoprotein A5 (APOA5) secreted by liver has an impact on liver immune microenvironment. To explore the function of APOA5 in SALI and the mechanisms of how it affecting liver immune, total RNA was isolated from livers of WT and Apoa5-/- mice treated with or without LPS using TRIzol reagent (n=5-6). The RNA integrity was assessed by Bioanalyzer 2100 with RIN number >7.0. Then, RNA sequencing was performed using an illumina Novaseq 6000. It is identified 316 consensus differentially expressed genes (DEGs) shared between LPS-challenged vs. untreated control and Apoa5-/-+LPS vs. WT+LPS group.
