RNA-seq was performed to compare the transcriptional programmes of murine Eµ-Crlf2/Jak2R683G-expressing B-ALL cells subjected to short-term (2 days) versus long-term (21 days) shRNA-mediated Jak2 knockdown in vivo. Overall design: NSG recipient mice of Eµ-Crlf2/Jak2R683G-pREBIR-shRenilla.713 or Eµ-Crlf2/Jak2R683G–pREBIR-shJak2.3323 cells were sacrificed at either 48 hr (acute knockdown) or 21 days (chronic knockdown) post shRNA induction and RNA-Seq was performed using total RNA isolated from dsRed+/eBFP2+ splenocytes. Experiment was performed in triplicate (three independent mice).
