(A) 诱导多能干细胞(Induced Pluripotent Stem Cell, IPSC)衍生的神经前体细胞(Neural Progenitor Cell, NPC)向神经元、星形胶质细胞(Astrocyte, Ast)及少突胶质细胞(Oligodendrocyte, OLs)等不同脑细胞类型的分化实验方案。(B) 分化流程结束后,各类脑细胞的相差显微镜成像结果(比例尺:100 μm)。(C) 对照细胞中,针对神经元(阳性免疫染色标志物为NeuN与β-Tubulin)、少突胶质细胞(阳性免疫染色标志物为MBP)及星形胶质细胞(阳性免疫染色标志物为GFAP)的代表性阳性免疫染色(作为分化效率标志物)与阴性免疫染色(作为细胞培养纯度标志物)结果。(D)-(E) 肾上腺脊髓神经病(Adrenomyeloneuropathy, AMN)与儿童型脑型肾上腺脑白质营养不良(childhood cerebral adrenoleukodystrophy, cALD)来源细胞中,针对神经元标志物(β-Tubulin)、星形胶质细胞标志物(GFAP)及少突胶质细胞标志物(MBP)的代表性阳性免疫染色结果。
