We compared gene expression among central memory (CD62L+CD127+; pop1), effector memory (CD62L-CD127+; pop2), and terminal effector (CD62L-CD127-; pop4) isolated from a single-cell-derived WT1-specific CD4+ T cell clone. Overall design: A CD4+ T cell clone was expanded by the stimulation with WT1 peptide and IL-2. The expanded cells were rested for O/N in IL-2-free medium and then separated into the 3 fractions on the basis of CD62L and CD127 expression by FACSAria. Gene expression was compared among the three populations in order to identify genes responsible for memory or effector differentiation and their maintenance.
