Head and neck squamous cell carcinoma (HNSCC) is one of the most common cancers globally, with the subtype affecting the oral cavity (OSCC) representing 40% of all head and neck cancers. With approximately 800,000 new cases and 400,000 related deaths annually, OSCC ranks as the sixth most prevalent cancer worldwide. The tumour microenvironment in OSCC forms a complex ecosystem that supports and protects the tumour, with dysregulation in cell-to-cell and cell-to-extracellular matrix (ECM) interactions being key contributors to cancer progression. In this study, we investigated the mechanistic roles of the Notch signaling pathways in sustaining the malignant phenotype. Using bulk RNA sequencing, we analyzed transcriptomic changes in SCC25 cell lines after exposure to a pan-Notch pathway inhibitor CB103.
